Endocannabinoids such as 2-arachidonoyl glycerol (2-AG) and arachidonoyl ethanolamide are biologically active lipids that are involved in a number of synaptic processes including activation of cannabinoid receptors. JW 642 is a potent inhibitor of monoacylglycerol lipase (MAGL) that displays IC50 values of 7.6, 14, and 3.7 nM forinhibition of MAGL in mouse, rat, and human brain membranes, respectively. JW642is selective for MAGL, requiring much higher concentrations to effectively inhibit fatty acid amide hydrolase activity (IC50s = 31, 14, and 20.6 µM for mouse, rat, andhuman brain membranes, respectively).

In laboratory procedures,for long term storage,we suggest that JW 642be stored assupplied at -20℃. It should be stable for at least one year.
JW 642 is supplied as a solution in methyl acetate. To change the solvent,simply evaporate the methyl acetate under a gentle stream of nitrogen and immediately addthe solvent of the choice. Solvents such asethanol,DMSO,and dimethyl formamide purged with an inert gas can be used. The solubility of JW 642 in these solvents isapproximately 13,5,and 11mg/ml,respectively.
JW 642 is sparingly soluble in aqueous buffers. For maximum solubility in aqueousbuffers,the ethanolic solution of JW 642should be diluted with the aqueous buffer ofchoice. JW 642 has a solubility of approximately 0.3 mg/ml in a 1:2 solution of ethanol:PBS(pH 7.2) using this method. We do not recommend storing the aqueous solution for more than one day.

4.How JW 642 works?

JW 642 is a potent and selective monoacylglycerol lipase (MAGL) inhibitor (IC = 3.7 nM). Displays >1000-fold selectivity for MAGL over fatty acid amidehydrolase (IC = 20.6 μM). Monoacylglycerol lipase (MAGL) isa serine hydrolase responsible for the hydrolysis of 2-AG to arachidonic acid and glycerol, thus terminating its biological function. Toxicological Effects may be caused like below: Methyl Acetate – Toxicity Data: Oral LD50 (rat): >5,000 mg/kg; Oral LD50 (rabbit): 3,705 mg/kg; Skin LD50(rabbit): >5,000 mg/kg; Inhalation TCLO (human): 15,000 mg/m3; Methyl Acetate – Irritation Data: Skin (rabbit): 500 mg (24h) mild; Eyes (rabbit):100 mg (24h)moderate.
